A study published in PNAS reveals that the metabolite AM404, derived from paracetamol, blocks pain by inhibiting voltage-gated sodium channels, NaV1.7 and NaV1.8, in peripheral sensory neurons. Experiments showed that AM404 suppressed channel activity and exhibited analgesic effects in in vivo tests, supporting the potential for developing targeted pain therapies.
