A novel 3D human epidermal equivalent model revealed that vitamin C treatment at concentrations of 0.1 mM and 1.0 mM led to significant increases in epidermal thickness and cell proliferation over 7 and 14 days. Changes in DNA methylation were highlighted by elevated 5-hydroxymethylcytosine levels, indicating a key epigenetic role for vitamin C. A total of 393 genes were differentially expressed, with implications for skin health and aging, suggesting vitamin C's potential therapeutic uses in reversing epidermal thinning.
